The Changes in Collagen Synthesis and Dermis Thickness
after in vivo Cryopreserved FatTransplantation

Abstract
The purpose of this study was to find out the change of skin structure after being grafted the cryopreserved fat(at the temperature of the -20¡É) tissue.
Human adipose tissue obtained by Coleman methods was refrigerated at the temperature of the -20¡Écorrectively for 8 weeks. Ten 8-week-old male nude mice were used in experiment.
In cryopreserved experimental group, all 10 mice were grafted 1.0 mL
cryopreserved fat tissue subcutaneously into left back. In the fresh control group, all mice were not grafted anything on right back. Eight weeks later, biopsies of the skin and subcutaneous tissue were performed and specimens were analyzed by histologic examination using hematoxylin-eosin stain. Dermal thickness was measured. The collagen fibers were investigated by immunohistochemistry. The growth factors such as PDGF & TGF-¥â1 affecting the collagen synthesis were measured by Real time RT-PCR.
Dermal thickness after cryopreserved fat grafting was significantly greater. On
cryopreserved fat grafting group,the synthesis of collagen type I and growth factors such as PDGF & TGF-¥â1 were significantly greater than control group.
The results of this experiment would be helpful for the study of the grafting with
cryopreserved fat tissues and to investigate to improvement of skin quality after fat grafting.
However, the long-term effects remain undetermined and need further investigation.
